چکیده

In recent years, there has been a growing interest in the formation of copolymer-lipid hybrid self-assemblies, which allow combining and improving main features pure lipid-based copolymer-based systems known for their potential applications biomedical field. As most common method used to obtain giant vesicles is electroformation, so far low Tg polymers flexibility at room temperature. Copolymers have synthesized by modified version ATRP, namely Activators ReGenerated Electron Transfer ATRP characterized NMR DSC. Giant obtained using electroformation droplet transfer method. Confocal fluorescence microscopy was image vesicles. Electroformation enabled narrow range compositions (15 mol% maximum copolymer content). This could be extended use method, obtaining incorporating methacrylate-based polymer wide compositions. Proof composition labeled lipids copolymers. work describes first time, best our knowledge, polymer/lipid formed with such content polymethylmethacrylate copolymer, glass temperature above shows that structures, more complex than ones mostly employed, can possibly included will give easier access functionalized stimuli-responsive exhibiting tunable permeability, these being relevant biological technological applications.

Asymmetric Hybrid Polymer–Lipid Giant Vesicles as Cell Membrane Mimics

Lipid membrane asymmetry plays an important role in cell function and activity, being for instance a relevant signal of its integrity. The development of artificial asymmetric membranes thus represents a key challenge. In this context, an emulsion-centrifugation method is developed to prepare giant vesicles with an asymmetric membrane composed of an inner monolayer of poly(butadiene)-b-poly(eth...

Hybrid, Nanoscale Phospholipid/Block Copolymer Vesicles

Hybrid phospholipid/block copolymer vesicles, in which the polymeric membrane is blended with phospholipids, display interesting self-assembly behavior, incorporating the robustness and chemical versatility of polymersomes with the softness and biocompatibility of liposomes. Such structures can be conveniently characterized by preparing giant unilamellar vesicles (GUVs) via electroformation. He...

Giant phospholipid/block copolymer hybrid vesicles: mixing behavior and domain formation.

Lipids and block copolymers can be individually assembled into unsupported, spherical membranes (liposomes or polymersomes), each having their own particular benefits and limitations. Here we demonstrate the preparation of microscale, hybrid "lipopolymersomes" composed of the common lipid POPC (1-palmitoyl-2-oleoyl-sn-glycero-3-phosphatidylcholine) and the commercially available copolymer PBd-b...

Modeling Enzymatic Pathways in Giant Lipid Vesicles

Giant vesicles (GVs) are artificial chemical systems largely used as cell models [1-3], since they are micrometer-sized closed compartments bounded by a semi-permeable membrane, which in turn is composed by self-assembling amphiphiles. By opportunely engineering the lipid membrane and by entrapping cascade-reaction enzymes inside GVs, it comes possible to design micro-sized reactors where metab...

Making giant unilamellar vesicles via hydration of a lipid film.

This unit describes protocols for making giant unilamellar vesicles (GUVs) based on rehydration of dried lipid films. These model membranes are useful for determining the impact of membrane and membrane-binding components on lipid bilayer stiffness and phase behavior. Due to their large size, they are especially amenable to studies using fluorescence and light microscopy, and may also be manipu...
